Cụ thể, mỗi tuần sẽ có 5 chuyến bay đi/về Cần Thơ – Phú Quốc vào các thứ 2, 3, 4, 5 và 7. Trong đó, chặng Cần Thơ – Phú Quốc khởi hành lúc 10h40, chặng Phú Quốc – Cần Thơ khởi hành lúc 12h15, các chuyến bay đi/về mất hơn một giờ bay, tùy thuộc vào điều kiện thời tiết.
Vietnam Airlines là hãng hàng không khai thác đường bay này với loại tàu bay ATR 72, sức chứa khoảng 72 hành khách.
Hồi tháng 11/2023, Bamboo Airways là hãng hàng không duy nhất khai thác đường bay Cần Thơ – Phú Quốc đã rút khỏi đường bay này cho đến nay.
